Analyte InformationEngineered chimeric human-murine antibody derived from the murine anti-beta-glucan mAb 2G8. The chimeric IgG was produced by fusing VH and VL regions of murine mAb 2G8 to the constant sequences of a human IgG1 gamma and lambda chain, respectively. The genes were recombinantly expressed using an Agrobacterium tumefaciens-mediated transient expression system in Nicotiana benthamiana plants. The recombinant chimeric 2G8 IgG antibody retained the antifungal activities of the original murine mAb 2G8 against Candida albicans.

ProtocolAfter blocking arrayed slides with 0.02% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), chimeric 2G8 IgG was assayed at 0.5 ug/ml, followed by biotinylated anti-human IgG (Vector) 1:200. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
