Analyte InformationMurine hybridoma IgM antibody, produced by Balb/c mouse immunization with a glyco-conjugate vaccine containing a soluble Candida albicans beta-glucan preparation (GGZym) conjugated to the recombinant, genetically inactivated diphteria toxin CRM197. mAb 1E12-IgM was shown to bind to the cell wall of fungal pathogens (Candida albicans, Aspergillus fumigatus and Cryptococcus neoformans) but did not confer protection in an experimental model of C. albicans infection.

ProtocolAfter blocking arrayed slides with 0.02% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), mAb 1E12-IgM was assayed at 0.2 ug/ml, followed by biotinylated anti-mouse IgM (Sigma) at 10 ug/ml. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
