Analyte NameA/Hamburg/5/09-E (222G), H1N1 virus
Analyte FamilyViruses/Virus-like particles
Analyte InformationBeta-propiolactone inactivated viruse derived from A/Hamburg/5/09 by egg passage and plaque purification in MDCK cells and differed by a single substitution, haemagglutinin D222G.
Concentration100 HA units
ProtocolAfter blocking arrayed slides with 3% (w/v) bovine serum albumin (Sigma A8577), 0.02% Blocker Casein (Pierce) in 150mM sodium chloride and 5 mM calcium chloride, the suspension of the inactivated virus (100 HA titre) was assayed in the presence of 17.5 uM oseltamivir carboxylate (19C for 1 h). After fixation with 4% paraformaldehyde, bound viruses were detected using the rabbit anti-A/New Jersey/76 antiserum (1:200), followed by biotinylated Protein A (Sigma), 10 ug/ml, and Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml). As diluent the blocker solution was used.
Other CommentsPlease note that the HA titre was measured under a different assay set up compared to that in publication PMID19741625. 2000 HA here equals to 100 HA in the abovementioned paper.
