ProtocolAfter blocking arrayed slides with 0.04% (v/v) Blocker Casein (Pierce), 1% (w/v) bovine serum albumin (Sigma A8577) in Hepes buffered saline (5 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM CaCl2), 2G12 was precomplexed with biotinylated anti-human IgG (Vector) at a 1:3 ratio (w/w) before application onto the slides at a final concentration of 10 ug/ml. Binding was detected using Alexa Fluor-647-labeled streptavidin from Molecular Probes (1 ug/ml).
